For the biotin labeled peptides, we often link biotin to the free -NH2 at N-terminal or Lys. The reaction is between the free -COOH of biotin and free -NH2 of peptide. As a matter of fact, there is no guanidine salts or DMSO introduced in the process of synthesis or modification of peptide. The guanidine salts and DMSO are often used for dissolve the hydrophobic peptides. The purification of peptides we usually use for antigen is >85% in our antibody production team so the byproduct of peptide synthesis and conjugation may not toxic ingredients.
