Tm value calculation is based on the primer concentration of 200 nM and the theoretical salt concentration of 50 mM. In the actual experiment, the primer or salt ion concentration has a great influence on the Tm value. Therefore, the Tm value calculation cannot be guaranteed to be completely consistent with the actual situation, and can only provide a reference for the experimental parameter setting.
