Our default vector for gene synthesis is pUC57. The difference between pUC57 and pUC57 simple vector is that pUC57 simple doesn't have the MCS. pUC57 simple vector has two commonly used sites: NdeI and EcoRV enzyme sites. Normally, for this vector, the gene will be cloned to EcoRV. If you choose NdeI/HindIII as the cloning site, you can use pUC57. The NdeI and HindIII on pUC57 are unique. Alternatively, Nde-HindIII sites can be synthesized additionally at both ends of the gene, and then insert it into the pUC57 simple vector.
